Study On The Expression Of Type ¢ñ And ¢ó Collagen And Matrix Metalloproteinase-1 In The Levator Ani Muscle During Late Pregnancy

Abstract
Objective To explore the expression levels of type I and type III collagen and matrix metalloproteinase-1 (MMP-1) mRNA in the levator ani muscles of late pregnant women and nulliparous women and the correlation between them. Methods 20 pregnant women in late pregnancy [age (28¡À4) years, gestational age (39¡À3) weeks] were selected as the late pregnancy group. At the same time, 5 married and childless women [age (25¡À4) years old] with perineal laceration who had levator ani muscle tissue samples collected due to perineal laceration were selected as the control group. HE staining was used to observe the morphology of the levator ani muscle, and immunohistochemistry and semi-quantitative reverse transcription PCR (RT-PCR) were used to detect the expression levels of type I and III collagen and MMP-1 mRNA in the specimens. Results The muscle fibers of the levator ani muscle tissue in the late pregnancy group were hypertrophic, tightly arranged but slightly disordered, and showed no signs of degeneration and necrosis. The expression of type ¢ñ and type ¢ó collagen and their ratio in the late pregnancy group were significantly higher than those in the control group (P<0.05); while the expression level of MMP-1mRNA was significantly lower than that in the control group (P<0.05). Conclusion Collagen synthesis in the levator ani muscle of women in late pregnancy increases, degradation decreases, muscle fiber hypertrophy occurs, and supporting strength increases.
