Abstract
¡¾Objective¡¿To perform complex enzyme hydrolysis on velvet antler collagen polypeptide, optimize the hydrolysis process, and determine the molecular weight distribution of the hydrolyzate. [Method] Using alkaline protease and trypsin as the test enzymes, selecting enzymatic hydrolysis time, pH value, enzymatic hydrolysis temperature, and enzyme addition amount as influencing factors, a response surface experiment design was used to optimize the single enzyme hydrolysis conditions. According to the response surface experiment The optimal conditions for single-enzyme hydrolysis were obtained to determine the dual-enzyme composite hydrolysis scheme. The molecular weight distribution of velvet antler collagen polypeptides was determined by Sephadex G25. [Results] The optimal hydrolysis conditions of alkaline protease are: enzymatic hydrolysis time 3.6 h, pH value 10.50, enzymatic hydrolysis temperature 55 ¡æ, enzyme addition amount 4%, hydrolysis degree of hydrolyzate is 22.03%; optimal hydrolysis conditions of trypsin are : Enzymatic hydrolysis time is 3.2 h, pH value is 8.00, enzymatic hydrolysis temperature is 50¡ãC, enzyme amount is 4.4%, and the degree of hydrolysis of the hydrolyzate is 15.81%. Compound enzyme hydrolysis conditions: enzymatic hydrolysis temperature is 52.5 ¡æ, adjust the pH value to 10.50, add 4% alkaline protease for enzymatic hydrolysis for 3.6 h; adjust the pH value to 8.00, add 4.4% trypsin for enzymatic hydrolysis for 3.2 h, and the resulting hydrolyzate The degree of hydrolysis can reach 33.42%. The molecular weight distribution of the collagen polypeptide hydrolyzed by the complex enzyme ranges from 400 to 1,400 u. [Conclusion] The process conditions for the compound hydrolysis of velvet antler collagen polypeptide by alkaline protease and trypsin were determined.
