Study On The Preparation And Activity Of Antioxidant Peptides From Tilapia Skin Collagen By Three-Enzyme Method

Abstract
In order to explore the best process for preparing tilapia skin collagen antioxidant peptide, based on the enzymatic hydrolysis of alkaline protease and trypsin, the enzymatic hydrolysis temperature, enzymatic hydrolysis time, enzymatic hydrolysis pH value and enzyme and substrate quality were used. The ratio ([E]/[S]) was used as the experimental factor, and the hydrolysis degree and DPPH free radical scavenging rate were used as response values. Response surface analysis was used to optimize the enzymatic hydrolysis conditions of Orientase 20A enzyme. The results show that the optimal process parameters are: enzymatic hydrolysis temperature of 41.74¡ãC, pH 3.97, enzymatic hydrolysis time of 6 hours, [E]/[S] of 1.5%, prediction of hydrolysis degree of enzymatic hydrolysis solution and DPPH free radical scavenging rate The values are 9.42% and 35.01% respectively, and the verification values are 9.57% and 35.21% respectively. The optimization of enzymatic hydrolysis conditions for the preparation of tilapia skin collagen antioxidant peptides by enzymatic hydrolysis of response surface is feasible and can be used for practical prediction. Antioxidant activity experiments show that the enzymatic peptide has strong ability to scavenge DPPH free radicals and ABTS+¡¤, with IC50 values of 10.78mg/mL and 8.26mg/mL respectively.
