Abstract
Konjac glucomannan (kgm) microspheres are used as the matrix, the kgm microspheres are activated with 1,4-butanediol diglycidyl ether, collagen is coated on the microspheres, and the collagen coating is cross-linked again , to obtain microcarriers with uniform and stable coating. Through the orthogonal regression combination experimental design of four factors and three levels, the effects of activation time, protein dosage, coupling time, and cross-linking agent dosage on the microcarrier cell culture effects were investigated. Taking the vero cell culture effect as an indicator, the optimal process for preparing collagen-coated microcarriers is activation time 5h, protein dosage 1:0.1 (ball:protein), coupling time 5h, and cross-linking agent dosage of 0.5ml per 1gkgm. agent. Under optimal preparation conditions, the maximum cell density of cultured vero cells can reach 1.7¡Á106 cells/ml, proving the feasibility of collagen-coated kgm microspheres as microcarriers for animal cell culture.
