Abstract
At low temperature of 4¡ãC, the collagen from the skin of artificially cultured giant salamanders was extracted using acid method (0.5 mol/L acetic acid) and enzymatic method (pepsin). Two different types of collagen were extracted and purified – ¡ªThe physical and chemical properties of acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC) were studied. The results show that ASC and PSC both contain 2 ¦Á chains and their dimer ¦Â chains, which are type I collagen; ASC and PSC have the highest glycine content, followed by glutamic acid and proline, and the lowest cystine content. ; The imino acid contents in ASC and PSC are 144 and 173 (based on residues/1 000 residues) respectively, and their thermal denaturation temperatures are 23.5 ¡æ and 26.5 ¡æ respectively, indicating that PSC has better thermal stability; ASC and PSC The maximum UV absorption peaks of ASC are located at 233.0 nm and 232.0 nm wavelengths respectively, which is consistent with the characteristics of collagen. The infrared characteristic absorption frequencies of ASC and PSC are similar, and both contain amide II band, amide III band and a series of absorption between the two amide bands. peak, indicating that the triple helix structure of collagen in ASC and PSC is relatively complete
