Study On The Properties Of Fish Skin Collagen By Circular Dichroism And Chromatography

Abstract
Acid-soluble collagen (asc) was extracted from carp (cyprinuscarpio) and cod (gadusmorhua) skins, and the properties of the two collagens were studied and compared. The denaturation temperature of carp skin ASC analyzed by circular dichroism (CD) is 36¡ãC, and the denaturation temperature of cod skin ASC is 19.0¡ãC, which is consistent with the analysis results of collagen imino acid content. Column chromatography results show that both collagens are composed of three different ¦Á chains (¦Á1, ¦Á2, ¦Á3), and the polymers formed by the three single chains contain two different forms: (¦Á1)2¦Á2 and ¦Á1¦Á2¦Á3. Pepsin enzymatic digestion profiles of the ¦Á1 chain of carp and cod skins showed obvious differences between the two collagens.
