Abstract
The intron sequence of human type I collagen ¦Á1(¢¡) chain (colIa1) gene has different transcriptional regulatory activities in different cells. It is reported that different segments of intron I of human col¢¡a1 gene (+544¡«+855 and +820 ~+1093) recombinant plasmids pscep-cat and pscip-cat were constructed and transfected into human embryonic tendon fibroblasts and tca8113 tongue cancer cells. The digoxigenin-labeled anti-cat-elisa test results showed that: pscep-cat was expressed in both cells were all expressed; pscip-cat was not expressed in human fibroblasts, but its expression in tongue cancer cells was significantly higher than that of pscep-cat. This indicates that the human col¢¡a1 gene intron ¢¡+544¡«+855 segment enhances chloramphenicol Acetyltransferase (cat) expression; the +820~+1093 segment inhibits human fibroblasts but significantly enhances cat expression in tongue cancer cells.
