Abstract
Abstract Objective To evaluate the safety of different concentrations of mitomycin C (MMC) collagen membranes used in non-penetrating trabecular surgery. Methods To establish a high-performance liquid chromatography (HPLC) method to determine the vitreous concentration of MMC in rabbit eyes, using centrifugation French-made porcine scleral dry collagen membranes were soaked in MMC solution to make MMC collagen membranes of different concentrations. Healthy purebred Japanese long-eared white rabbits were randomly divided into 6 groups and underwent non-penetrating trabecular surgery on both eyes. A, B, The four groups C and D were jointly implanted with MMC collagen membranes of different concentrations. Group E used 200 mg/L MMC once during the operation, and group F was the blank group. The corneoscleral limbus conditions in the surgical area were compared 1 week after operation. HPLC method was used to detect and compare the MMC concentration in the vitreous cavity of each group at 4 weeks. Results Under this chromatographic condition, the linear range of MMC in the vitreous cavity was 0.1 to 2.5 mg/L, and the correlation coefficient (r) was 0.9970. After surgery At 1 week, the MMC concentration in groups C and D was significantly higher than that in group E (P<0.001); at 4 weeks after surgery, there was no significant difference between group A and group E (P>0.05). Higher than that of group E (P<0.01). Conclusion The concentration of MMC in the vitreous cavity increases with the increase in the concentration of MMC released by the collagen membrane. The concentration of MMC in the vitreous cavity of the collagen membrane loaded with low concentration MMC (50 mg/L) is safer.
