Study On The Technological Conditions For Enzymatic Extraction Of Squid Skin Collagen

Abstract
The process conditions for enzymatic extraction of squid skin collagen were studied. Based on the content of hydroxyproline in the hydrolyzate of five proteases, two enzymes with high extraction rates, trypsin and papain, were determined to be hydrolysis enzymes. The l16(45) orthogonal test was used to determine the hydrolysis of squid by these two enzymes. skin to prepare optimal enzymatic hydrolysis conditions for collagen. The results show that the optimal temperature, enzyme amount, substrate concentration, ph, and time for trypsin and papain to hydrolyze squid skin to prepare collagen are 55¡ãC, 1200u¡¤g-1, 1:20, ph8.0, respectively. 4h and 50¡æ, 3200u¡¤g-1, 1:20, ph6.0, 6h. The collagen contents extracted by the two proteases were 11.08% and 11.36% respectively, and the extraction rates were 95.16% and 97.56% respectively.
