Abstract
Objective To study the therapeutic effect and mechanism of Zixue Powder on collagen-induced arthritis in rats. Methods Wistar rats were randomly divided into 5 groups, namely control group, model group, indomethacin tablets (2 mg/kg) group, and Zixue powder 0.5 and 1.0 g/kg groups, with 10 rats in each group. Establish a rat type ¢ò collagen-induced arthritis model. Rats were administered intragastrically on the 12th day after the initial immunization, with a dosage of 10 mL/kg, once a day. Both the control group and the model group were given the same volume of normal saline by gavage. Each group continued to be administered for 3 weeks. The width, thickness and circumference of the left hind ankle of the rats were measured, and the arthritis index was calculated. Observe the pathology of rat ankle joints. The serum inflammatory factor platelet-activating factor (PAF) level, cyclooxygenase-2 (COX-2) activity, and tumor necrosis factor ¦Á (TNF-¦Á) levels were measured using ELISA, and serum circulating immune complexes ( CIC) level. Results After 7 days of administration, the ankle joint width and thickness of the Zixue Powder group were significantly lower than those of the model group, and this continued until the 21st day of administration. Synovial cell hyperplasia and synovial tissue congestion and edema were significantly reduced in the Zixue Powder group. Compared with the model group, the systemic performance score, joint swelling index and arthritis index of the Zixue Powder group were significantly reduced (P<0.05, 0.01, 0.001). Compared with the model group, the serum PAF level and COX-2 activity in the Zixue Powder 0.5 and 1.0 g/kg groups were significantly reduced (P<0.001). Compared with the model group, the serum TNF-¦Á and CIC levels in the Zixue Powder 0.5 and 1.0 g/kg groups were significantly reduced (P<0.05, 0.01, 0.001). Conclusion Zixue Powder has a therapeutic effect on collagen-induced arthritis damage in rats, and its mechanism may be related to the anti-inflammatory and antioxidant effects of Zixue Powder For further information of this article and research, feel free to contact our team for asssitance.
