Abstract

Abstract Purpose: To explore the effect of TGF¦Â1/Smad3 signaling pathway on changes in lysyl hydroxylase 2 (LH2) activity, as well as the role and mechanism of this change in collagen deposition in pulmonary fibrosis. Methods: The human lung fibroblast cell line HFL1 was cultured in F12 medium containing 10% fetal bovine serum. The experiment was divided into a control group, a TGF¦Â1 (10 ¦Ìg/L) stimulation group and a minoxidil (5 ¦Ìmol/L) intervention group. The same amount of culture medium was added to the control group; RNA and protein from each group were collected after 48 h, and RT- qPCR was used to detect the mRNA expression levels of PLOD2, ¦Á-SMA and COL ¢ñ, Western blot was used to detect changes in protein levels of LH2, total Smad3, phosphorylated Smad3, ¦Á-SMA, COL ¢ñ and COL ¢ô, and ELISA was used to detect hydroxylysylpyridinol in cells. Results: After TGF¦Â1 stimulation, the mRNA expressions of PLOD2, ¦Á-SMA and COL¢ñ were up-regulated (P<0.01); their corresponding protein levels such as LH2, phosphorylated Smad3, ¦Á-SMA, COL¢ñ and COL¢ô protein levels increased, while total Smad3 There was no change in protein levels. The mRNA and protein expression of the above genes decreased after minoxidil intervention (P<0.01). After TGF¦Â1 stimulation, ELISA detected an increase in HP content compared with the control group, and the synthesis was reduced after minoxidil intervention (P<0.05). Conclusion: The activation of the TGF¦Â1/Samd3 signaling pathway increases the expression of LH2; minoxidil reduces the expression of LH2 and the synthesis of hydroxylated collagen pyridine chains by inhibiting TGF¦Â1/Samd3 signal transduction, thereby alleviating pulmonary fibrosis.
