Abstract
In order to study the role of human chymase (hChymase) in the formation of cardiac angiotensin II (AngII) and its significance in the myocardial remodeling process, MLC2-human chymase transgenic mice were established. RT-PCR method was used to analyze The tissue-specific expression of chymase gene and the transcriptional expression of type ¢ñ and ¢ó collagen genes in the heart were studied, and the activity of cardiac chymase and angiotensin converting enzyme (ACE) as well as the Ang II content in heart and plasma were detected by radioimmunoassay. , the gelatin zymography method was used to detect cardiac matrix metalloproteinase (MMP) activity; and the cardiac hypertrophy phenotype of chymase transgenic mice was preliminarily observed using physiological and morphological methods. And non-transgenic mice (control group) ) comparison, the results show that (1) the chymase transgene can be specifically expressed in mouse hearts; (2) the chymase activity in the heart of chymase transgenic mice is higher than that of the control ((0.27¡À0.07)U/mgvs (0.15¡À0.02)U/ mg, Pvs (0.18¡À0.02) U/mg); (3) Ang II content in the heart of chymase transgenic mice increased, about 3 times that of the control ((1984¡À184) pg/g vs (568¡À88) pg/g ,PP
