Abstract
Objective To explore the expression of matrix metalloproteinase 3 (MMP-3) in the supraspinous ligament of ankylosing spondylitis (AS) and its role in ligament degeneration. Methods Immunohistochemistry was used to detect the expression of MMP-3 and type I collagen in the ligament. Enzymelinked immunosorbent assay (ELISA) was used to detect the content of proteoglycan in the ligament. Scanning electron microscopy and transmission electron microscopy were used to examine the supraspinal ligament. Morphological observations. Image-ProPlus (IPP) quantitatively analyzed the distribution density of collagen fibrils and the percentage of collagen-positive staining area. Results MMP-3 was highly expressed in ligament vascular smooth muscle cells and endothelial cells in the study group, but no expression was found in the control group. The proteoglycan content in the control group was (19.462¡À2.174) ¦Ìg/mg, and in the study group was (7.477¡À2.512) ¦Ìg/mg. ; Under the electron microscope, the arrangement of collagen fibers in the control group was regular, and the collagen fibrils in the study group were obviously exposed, and a large amount of matrix was degraded; the distribution density of collagen fibrils in the control group was (218.02¡À10.39)/¦Ìm2, and that of the study group was (122.47¡À22.42)/¦Ìm2; control The percentage of positive staining area for type I collagen in the group was (60.23¡À3.41)%, and that in the study group was (43.37¡À5.96)%. The difference between the two groups was statistically significant (P<0.05). Conclusion MMP-3 is highly expressed in the supraspinous ligament blood vessels of patients with active AS, which may participate in the pathological process of ligament degeneration and ossification by promoting blood vessel growth, degrading proteoglycans, and destroying the normal microstructure of the ligament.
