Abstract
¡¾¡¿Objective to explore the role of catecholamines (cas) derived from th17 and treg cells in the development and progression of collagen-induced arthritis (cia). Methods Eighteen male dba/1 mice were randomly divided into control group, cia model group I (35 days) and cia model group II (55 days). The cia mouse model was prepared by injecting type II collagen emulsion into the tail root. The real-time PCR method detects the specific transcription factor ROR-¦Ãt of Th17 cells in lymph nodes, the related cytokines IL-17 and IL-22, the specific transcription factor Foxp3 of Treg cells, the related cytokines transforming growth factor (TGF)-¦Â and mRNA expression of tyrosine hydroxylase (TH). Immunofluorescence histochemical staining was used to observe the co-localization of ror-¦Ãt and foxp3 with th, vesicular monoamine transporter-2 (vmat-2) and monoamine oxidase (mao) respectively in lymph nodes. Results Compared with the control group, the mRNA expression of ror-¦Ã, il-17, th and il-22 in the lymph nodes of mice in the cia model group ¢¡ and model ¢¢ group increased; the mRNA expression of foxp3 and tgf-¦Â decreased; among the models The mRNA expression of il-17 in group ¢¢ was decreased compared with model group ¢¡. There were co-localized cells of ror-¦Ãt and foxp3 with th, vmat-2 and mao respectively in the lymph nodes of mice in the control group, cia model group ¢¡ and model ¢¢ group. The number of ror-¦Ãt/th, ror-¦Ãt/vmat-2 and ror-¦Ãt/mao double-positive cells in the lymph nodes of mice in the cia model group increased. The number of these double-positive cells in the model group II was slightly higher than that in the model group I. reduce. Conclusion The ability of Th17 cells to synthesize cas in the lymph nodes of mice in the cia model group is enhanced. This increase in cas derived from th17 cells may have a certain anti-inflammatory effect in the occurrence and development of cia.
