Abstract
This article aims to comparatively study the morphological differences between endothelial cells and cardiac tissue in two-dimensional and three-dimensional culture modes. Various type I collagen three-dimensional culture models such as in-gel, on-gel, sandwich mode, and slide culture chamber models were used to observe the growth of tissues and cells through immunofluorescence technology and microscopic morphology. In two-dimensional plane culture, primary cardiac vascular endothelial cells are arranged like paving stones; in three-dimensional collagen culture mode, endothelial cells take on a long spindle shape and migrate into the collagen culture medium, and are involved in angiogenesis and angiogenesis in vivo. The endothelial cell activation phenotype during the process was similar. Adding vascular endothelial growth factor (VEGF) can enhance the formation of endothelial cell tubular structures. In the three-dimensional collagen, the heart tissue blocks grew well, the migrating cells connected adjacent tissue blocks, and the tissue blocks beat spontaneously. This work shows that improved thin-layer collagen culture, slide culture chamber model and arterial strip model are better tools for studying angiogenesis and angiogenesis. In the case of three-dimensional culture, endothelial cells can form tubular structures through spatial proliferation, migration, and anchoring, which are more suitable for the study of angiogenesis than two-dimensional flat culture. Different three-dimensional culture models can be used for research with different purposes.
