Three-Dimensional Parallel Collagen Scaffold Promotes The Formation Of Tendon-Like Extracellular Matrix

Abstract

Objective: To explore the effect of three-dimensional parallel collagen scaffolds on the morphology, arrangement and extracellular matrix formation of tendon-derived stem cells. Methods: One-way freezing technology was used to prepare three-dimensional parallel collagen scaffolds, and traditional freeze-drying technology was used to prepare three-dimensional disordered collagen scaffolds. By transplanting tendon-derived stem cells in vitro and subcutaneous scaffolding, the effects of different three-dimensional structures on cell morphology, arrangement and extracellular matrix formation were observed. Results: The three-dimensional parallel collagen scaffold showed a three-dimensional parallel structure. The cells planted on the three-dimensional parallel collagen scaffold showed spindle-like tenocyte morphology, and the cells were arranged along the fiber direction of the collagen scaffold, forming a dense cell layer on the seventh day; the cells on the three-dimensional disordered collagen scaffold showed multiple shape, randomly distributed on each collagen scaffold sheet. In the subcutaneous implantation experiment, fibroblast-like cells from the host migrated into the scaffold after 2 weeks. The three-dimensional parallel collagen scaffold was mainly composed of elongated fibroblast-like cells, and the cells were arranged along the direction of the scaffold; while the cells in the three-dimensional disordered scaffold were mostly Oval-shaped, and the distribution has no certain directionality. Conclusion: The parallel three-dimensional structure induces cells to become elongated, promotes cells to be arranged in parallel, and at the same time promotes the formed extracellular matrix to be distributed in parallel directions. The results of this study can provide reference and reference for the ideal scaffold construction for tendon tissue repair.
