Abstract
Objective To explore the role of type ¢ò collagen (collagen ¢ò) in promoting the directional differentiation of rat bone marrow mesenchymal stem cells (BMSCs) into chondrocytes in vitro. Methods: BMSCs were cultured in vitro and divided into different collagen II inoculation concentration groups (25, 50, 100, 200 ¦Ìg/mL) and blank control groups, which were treated on BMSCs for 7, 14, and 21 days respectively. The CCK-8 method was used to detect the effects of different concentration groups on the activity of BMSCs. Effect; qPCR and immunofluorescence methods were used to detect the differences in collagen II, SOX9 and aggrecan mRNA and protein expression between each concentration group and the blank control group after treating BMSCs in different concentration groups. Results ¢Ù CCK-8 test results showed that there was no statistically significant difference in 7, 14, and 21dD (450) values among each concentration group (P>0.05); ¢Ú qPCR test results showed that after 21 days, compared with the blank control group, collagen ¢ò and SOX9 and aggrecanmRNA expression levels increased (P<0.05), with the 100 ¦Ìg/mL group being the highest (P<0.01); on 14 days, the expression levels of collagen ¢ò, SOX9 and aggrecanmRNA in each concentration group increased? 100¦Ìg/mL? group was most obviously up-regulated (P<0.05?), but at 7 days, there was no statistically significant difference in the expression of mRNA among the three groups (P>0.05); ¢Û The immunofluorescence results of each concentration group at 21 days showed: at 100 ¦Ìg/ The expression levels of collagen II, SOX9 and aggrecan mRNA proteins in the mL¢ò group were significantly higher than those in the other groups (P<0.05). Conclusion Collagen¢ò can promote the differentiation of rat BMSCs into chondrocytes in vitro, among which? 100¦Ìg/mL? The group promotion effect is the most obvious.
