Abstract
Abstract Acid-soluble collagen (ASC) from Yunnan sea bream skin was extracted using low-temperature acid method. The physical and chemical properties of ASC such as amino acid composition, subunit composition, structural characteristics, thermal stability and solubility were studied to provide a basis for the preparation of Yunnan sea bream skin. Comprehensive utilization provides a theoretical basis. Research shows that the main amino acids of ASC are glycine, proline and alanine, with lower contents of tyrosine, methionine and cysteine, and the hydroxylation rate of proline is 35.47%; SDS-PAGE electrophoresis patterns show that ASC It is mainly composed of ¦Á1, ¦Á2 and ¦Â chains, which is consistent with the characteristics of type I collagen; the maximum UV absorption wavelength of ASC is 230 nm; infrared spectrum and X-ray spectrum show that ASC molecules are arranged regularly and compactly, with a complete triple helix structure; difference Calorimetric scanning showed that the thermal denaturation temperatures of ASC were 82.1 and 222.3¡ãC respectively, indicating that ASC has good thermal stability; scanning electron microscopy showed that ASC molecules have a three-dimensional multi-sheet structure and are evenly distributed, making it suitable for use in food, medicine and cosmetics. Carrier and biomedical base; ASC has good solubility at pH less than 4 or NaCl concentration less than 4% (w/v)
