Abstract
Objective: To study the effect of tilapia skin collagen peptide on the proliferation ability of human colon cancer cell line LoVo in vitro, and to preliminarily explore its mechanism of action. Methods: Human colon cancer LoVo cells were cultured in vitro, and the effect of tilapia skin collagen peptide on cell proliferation was detected by methylthiazolyldiphenyl-tertrazolium bromide (MTT) method; changes in cell membrane and nucleus were observed by fluorescent probe staining; laser confocal The expression of reactive oxygen species (ROS) in cells was observed under a microscope; the activities of mitochondrial respiratory chain enzyme complexes I and III were detected by colorimetry. Results: Tilapia skin collagen peptide had a certain inhibitory effect on the proliferation of LoVo cells. When the mass concentration was 100 mg/mL and the action was for 48 hours, the inhibition rate reached 55.03% (P<0.05); compared with the blank control group Ratio, after the action of collagen peptides, the nucleus of LoVo cells was concentrated, the cell membrane appeared incomplete, and the cells were apoptotic; the expression of intracellular reactive oxygen species was significantly increased (P<0.05), and the activity of intracellular mitochondrial respiratory chain complex III was significantly reduced (P <0.05), but has no effect on the activity of mitochondrial respiratory chain complex I. Conclusion: Tilapia skin collagen peptide has a certain inhibitory effect on the proliferation of human colon cancer cells, which may be related to inhibiting the activity of mitochondrial respiratory chain complex III, increasing intracellular ROS levels, and damaging the cell membrane morphology.
