Abstract
Objective To investigate the anti-tumor effect of anti-type IV collagenase monoclonal antibody 3g11 and pingyangmycin (pym) conjugate. Methods Polyglutamic acid (plg) was used as the intermediate carrier to prepare the 3g11-plg-pym conjugate, and its inhibitory effect on tumor cell proliferation was determined by the mtt method. The mouse transplanted liver cancer h22 was used as a model to observe the anti-tumor effect in vivo. Results: The conjugate retained the immunological activity of monoclonal antibody 3g11 against type IV collagenase, and its killing effect on H22 and KB cells cultured in vitro was weaker than that of pym. In animal experiments, the inhibitory rate of pym10mg¡¤kg-1 against h22 liver cancer was 60.6%, while the tumor inhibition rate of 3g11-plg-pym conjugate at an equal cytotoxic dose reached 90.8%, which can significantly prolong the survival time of mice compared with pym. bit survival time. Conclusion The 3g11-plg-pym conjugate has a stronger anti-tumor effect on mouse liver cancer h22 than pym, and may become a new anti-tumor targeted drug.
